Pairing Estrous Females and Sterile Males

1. Once the estrous females and stud males are ready for mating, cage them together overnight.

2. Check for vaginal plugs in the morning on day 4 and record that this is the first day of pregnancy.

Tips: If mating takes place, a white or yellow vaginal plug composed of male secretions from the vesicular and coagulating glands should be present within 12 h following mating. The plug may be visible at the surface or out of sight deep in the vagina. Therefore, a narrow, clean steel spatula or its equivalent (3 to 4 mm wide) may be used to detect the plug in the vagina, which feels harder than the surrounding soft tissue. Thus, pseudopregnancy has been induced because the males are sterile.
