Studies in female athletes have shown no effect on body composition or muscle strength following supplementation of 500 fjg chromium picolinate daily during 6 weeks of resistance training (Livolsi et al 2001). In a clinical trial of older women a high-dose chromium picolinate supplement did not affect body composition, skeletal muscle size or maximal strength above that of resistance training alone (Campbell et al 2002). A meta-analysis of trials of dietary supplements for enhancing lean muscle mass and strength during resistance training did not support the use of chromium for this purpose (Nissen & Sharp 2002).
Necessary for determination of lineages that are derived from the myotomes 20, 21 . In mice lacking both genes, no muscle precursor cells are generated. In contrast, myogenin and MRF4 are not required for skeletal muscle lineage determination, but are active downstream of the other two members of the MyoD family myogenin is responsible for fusion of muscle cells and the formation of myotubes, whereas MRF4 plays a role in the subsequent differentiation of myotubes into mature myofibers 12, 13, 15 . In adult muscle fibers, MRF4 is the predominant transcript 7 . Recent reviews on this topic, the regulation of expression of bHLH transcription factors and the role of integrins and cadherins in muscle development and migration, include references 22-25, all collected in a special issue of Cell and Tissue Research.
Trisomy 18, initially identified in 1960,83 causes serious birth defects including a low birth weight microcephaly a characteristic facies with fawnlike ears, micrognathia, high arched palate, a narrow bifrontal diameter, and prominent occiput hypertonicity poor muscle development hand flexion with overlapping of second and fifth fingers and limited hip abduction (Fig. 3-10). Rocker-bottom feet, webbing of toes, and dorsiflexion of a short great toe are common. Renal anomalies including horseshoe kidney and collecting duct malformations, cardiac malformations, pyloric stenosis, eventration of the diaphragm, and Meckel diver-ticulum may occur. Apnea and failure to thrive are serious complications. The majority (90 ) die before 1 year of age.
The study of sex differences in the brain grew out of our understanding of how gonadal hormones influence sexual differentiation of the body with regard both to primary sexual differences between males and females (the differentiation of the sexual organs) and to the development of secondary sexual differences (body fat distribution, muscle development, breast development, differences in hair distribution). In the case of sexual differentiation of the body, it is clear that exposure of males to various testicular secretory products, including testosterone, during early prenatal development leads to sexual differentiation of the internal and external genitalia. Later activation of the reproductive axis at puberty, with a sustained increase in circulating testosterone, then leads to the development of secondary sexual characteristics. Not all pathways are as deeply conserved as the Hox system, nor rely on conserved linkage relationships. Even this classic system is subject to all the sources of variation that one might expect in fact, the more intensely it is studied, the less completely conserved it seems to be (e.g., Levine et al. 2002), including the variety of ways the genes in the cluster may be regulated to coordinate their spatial and temporal domains of expression (Kmita and Duboule 2003). Nonetheless, much is conserved, and similar situations apply to a number of other differentiation hierarchies, such as the role of Pax6 in eye development (Chapter 14), or MyoD in muscle development (Gerhart and Kirschner 1997). Figure 7-4B shows the general logic of a selector gene, and the way that REs are used to start and proliferate a cascade of downstream effects on gene expression.

More precise estimates can be obtained by using the body surface mass-index, but for practical purposes, a 0.4-0.5 multiplier gives a reasonable approximation. The upper limits are equally significant, and are considered pathological, regardless of body size, if more than 350-400 gm in a woman, or more than 400-450 gm in a man. Basic helix-loop-helix (bHLH) TFs form dimers in each of two helical domains, that are separated by a loop DNA binding is by a region of 10-13 basic amino acids at the end of the first helix in each of the polypeptides. The two molecules may be of the same or from a related bHLH gene. bHLH genes are important in a variety of neural and sensory developmental functions, and include the MyoD genes used in muscle development.
