Somatic Mutations In Cloning

Somatic mutation and somatic recombination following DNA damage and repair could affect the outcome of cloning procedures. Exposure to mutagenic chemicals and radiation can obviously damage somatic cells. Nondividing stem cells within adult tissues could be less susceptible to damage by transient exposures. Cloning with somatic cells bearing de novo mutations would then give rise to cloned embryos displaying either lethality or aberrant phenotypes later in life. Recombination-based repair of DNA damage, as well as aberrant mitoses can lead to loss of heterozygosity or uniparental disomy for entire chromosomes or chromosome regions. Loss of heterozygosity could lead to phenotypic exposure of recessive traits, whereas uniparental disomy could lead to abnormal phenotypes associated with imprinted regions of the genome.
