In this chapter, we have detailed a method for the production and maintenance of human monoclonal T cell populations in long-term culture, and we have provided selected examples of protocols for analytic techniques with which to seek age-associated changes therein (biomarkers of aging). The model represents a situation of chronic antigenic stress, where TCC are constantly reexposed to stimulating antigen.
