Appropriate blanks are also run during the assay to correct for the positive fluorescence of the mitochondria themselves. These blanks have mitochondria and all the reaction components but do not contain substrate. Values are obtained by subtracting the fluorescence of the blanks from the fluorescence of the samples and dividing the results by the 15 minutes of incubation. Special blanks (also without substrate) must be included to correct for the fluorescence of antimycin A when this mitochondrial respiratory inhibitor is included in the reaction.
