Transgenic mouse models are being used to investigate cigarette smoke-induced emphysema.31 The cigarette-smoking protocol consists of placing mice in a polypropylene whole-body inhalation chamber containing inlet pores and an exhaust pore. Smoke is generated through a 50-mL glass syringe equipped with a cigarette holder from one or multiple unfiltered cigarettes. The exposure period usually is long term on a daily basis. A typical protocol is exposure 6 days per week, for 6 months. Mice typically demonstrate progressive inflammatory cell recruitment beginning within the first month of smoking followed by airspace enlargement after 3 to 4 months of cigarette exposure.
