Allergen Induced Asthma

Mice are primed intraperitoneally with 10 pg ovalbumin (OVA) and 20 mg Al(OH)3 in 0.2 mL of phosphate-buffered saline (PBS, pH 7.4) once per week for 3 weeks. Twenty-four hours after the last injection, the primed mice are placed individually in a 50-mL plastic tube and challenged by exposure to aerosolized 5% OVA/PBS, delivered by a nebulizer driven by compressed air at 5 L/min for 20 min. Challenge is carried out once a day for 6 consecutive days. Marked eosinophilic inflammation and AHR are generally induced by this treatment.29
