When the radiolabeled ligand binds to the receptor attached to the SPA bead, it will be in close proximity to the scintillant on the bead, generating photons, which can be measured in a TopCount or MicroBeta plate counter. When a compound competes with radiolabeled ligand in binding to the receptor, the radiolabel is displaced by the competitor and will have a reduced signal. The radiolabel, which is not bound to the SPA bead, will not be in close proximity of the scintillant and will not give any signal.
